PT - 141
PT-141 (Bremelanotide) is a synthetic cyclic peptide analog of alpha-melanocyte-stimulating hormone (alpha-MSH) studied for its central effects on the melanocortin system and sexual behavior. Unlike PDE5 inhibitors which act peripherally on vascular smooth muscle, PT-141 research focuses on direct central nervous system pathways affecting sexual motivation and arousal in preclinical models.
Mechanism of Action
• Activates melanocortin receptors (MC1R, MC3R, MC4R) in the hypothalamus and limbic system, studied for effects on sexual arousal pathways in animal models.
• Investigated for increases in dopaminergic activity in the mesolimbic system, influencing motivation-related neurobiology.
• Research suggests stimulation of nitric oxide synthesis in relevant neural circuits.
• Studies indicate activity regardless of vascular health status, distinguishing its mechanism from PDE5-based pharmacology.
Research Areas
• Hypoactive sexual desire disorder (HSDD) research; an FDA-approved indication exists for a subcutaneous formulation.
• Erectile dysfunction biology, including cases studied alongside or in comparison to PDE5 inhibitor mechanisms.
• Sexual dysfunction secondary to pharmacological interventions.
• Central arousal pathway and melanocortin system research.
